4.1.3. Protoplast isolation and culture

A few studies have explored date palm protoplast culture (Chabane et al., 2007; Rizkalla et al., 2007; Assani et al., 2011; Titouh et al., 2015, 2017). They have defined conditions for the recovery of protoplasts. Protoplasts from various genotypes have been recovered from leaves (Chabane et al., 2007; Rizkalla et al., 2007) and from embryogenic cultures (Chabane et al., 2007; Titouh et al., 2017) using mixtures of cell wall-degrading enzymes. The protoplasts can be efficiently isolated by passage through a metal filter and subsequently purified over a cushion of 21% or 25% sucrose (Titouh et al., 2015). Protoplasts have regenerated cell walls and microcalluses have been recovered. Protoplast culture of date palm could be used for somatic hybridization; however, the efficacy of this technology for date palm is not clear.
