5.1.4. Triploid recovery

Regeneration of triploid plants from endosperm was reported by Sun et al. (2011). Callus was initiated from immature endosperm on MS medium with 6.0 μM 2,4-D, 2.5 μM NAA and 4.0 μM kinetin after 21 days. Buds developed on media containing either BA or TDZ with NAA. Shoots elongated on medium containing 3 μM IAA (indole-3-acetic acid) and 1.5 μM BA; 90% of the shoots formed roots on semisolid ½ MS medium with 2 μM IBA. The triploid plants had larger stomata and more chloroplasts in the guard cells. More than 75% of endosperm-derived plants were triploids: 3n = 3x = 27. Triploid plants were also recovered from anther cultures of male papaya flowers (Rimberia et al., 2006) (see above); however, regenerants were all female.
