5.1.3. Haploid recovery

Haploid papayas were recovered from cultured anthers (Litz and Conover, 1978b, 1979; Tsay and Su, 1985). Rimberia et al. (2005, 2006) cultured anthers containing uninucleate, mitotic and binucleate pollen from male plants on semisolid MS medium with 0.04 μM CCPU (N-(2-chloro-4-pyridyl)-N′-phenylurea) and 0.5 μM NAA. Sex of regenerated plants was determined with DNA markers and their ploidy was estimated by flow cytometry. Regenerated plants were all female; 26 were triploid and one was tetraploid. No haploids were reported. Litz and Conover (1978b) reported haploids with n = x = 8 (the normal haploid number is 9).
