4.1.4. Protoplast isolation and culture

Reports on pineapple protoplast isolation are very limited. Guedes et al. (1997) isolated protoplasts from ‘Perolera’ using a combination of 2% meicelase, 2% rhozyme and 0.3% macerozyme supplemented with antibiotics, while Zhao et al. (2011) isolated protoplasts from ‘Josapine’ in a 5% mannitol solution with 1.5% macerozyme and 1.5% cellulase followed by gradient centrifugation. The latter authors reported a high protoplast yield (3.6 × 106/g FW) with 88.9% viability. No regeneration was reported.

Protoplast culture and somatic hybridization are likely to have limited impact on pineapple improvement. Although protoplasts have been used in other monocotyledons for genetic transformation, methods based on biolistics and Agrobacterium have proven more effective with pineapple.
