2.2. Gene cloning and proteomics

Оглавление

UniProtKB search indicates 95 entries related to A. occidentale. Of these, 49 gene sequences were related to photosynthesis, 8 barcoding loci sequences were related to ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit (rbcl) and maturase K (matK), 27 gene fragments of protein synthesis components, 4 sequences of seed storage proteins and 7 other sequences of unknown function are available in the public domain. Cashew nut allergy is the second most commonly reported tree nut allergy in the USA (Robotham et al., 2005). To address the allergen in cashew, its seed storage protein genes have been sequenced, expressed in Escherichia coli and followed by allergy testing. Wang et al. (2002) cloned and characterized major cashew allergen protein Ana o 1, a vicilin. Wang et al. (2003) isolated and tested a legumin-like seed storage protein Ana o 2, having allergen-like properties. Robotham et al. (2005) describe a third major cashew allergen, a 2S albumin, Ana o 3. They also reported a recombinant cashew 2S albumin gene which was amplified from a cDNA library by PCR, sequenced and expressed in E. coli for reduced allergy reaction.
