Considerable variation occurs among plantlets that develop from the rooted shoots that are regenerated from cotyledonary nodes without intermediate callus. Variation included differences in height, stem diameter, plagiotropic growth and leaf phenology. Ananthakrishnan et al. (2002) reported excessive variation in plants regenerated from adventitious shoots derived from cotyledonary segments of mature embryos. DNA methylation studies of the variants revealed the same polymorphic bands, suggesting the possibility of hotspots of DNA methylation. Such variation in phenotype needs long-term field studies because in vitro stress can cause transient variation in plants (Nivas and D’Souza, 2004).
