While PD is a complex and heterogeneous multisystem disorder [3], the cardinal motor features of PD, that is, the triad of akinesia/bradykinesia, tremor at rest, and muscular rigidity, remain its defining, and clinically most troublesome, set of features in mild-to-moderate cases of PD. These features result from decreased dopaminergic transmission in the motor portions of the basal ganglia, in particular the putamen, due to progressive loss of innervation from dopaminergic neurons in the SNc. The following description focuses on the circuit-level consequences of the loss of dopamine.
