T Cell-Engaging Therapies

Targeting known tumour-specific or tumour-associated antigens is also the main rationale of chimeric antigen receptor T cells and T cell-engaging bispecific antibodies. With these approaches, after binding the targeted antigen on the tumour cells, every T cell irrespective of its original antigen specificity and costimulatory signalling will be able to exert cytolytic function against the cells it is bound to [33, 34]. This approach has shown major success in haematological malignancies, especially CD19+ B cell leukaemia [35]. The major obstacle in solid tumours is the immunosuppressive tumour microenvironment blocking the T cells from entering the bulk of tumour cells.
