2.3. Gene cloning

Manganese superoxide dismutase (MnSOD) was successfully cloned and expressed in Escherichia coli to determine if pistachio MnSOD could cause allergic reactions in atopic subjects (Noorbakhsh et al., 2010). The results showed that this protein might trigger some cross-reactions to IgE antibodies and thus could be considered as a panallergen. In another study, a PCR approach with degenerate primers that were designed according to conserved nucleotide binding site–leucine-rich repeat (NBS-LRR) regions of known disease-resistance (R) genes was used to amplify and clone homologous sequences from P. atlantica subsp. kurdica. The isolated NBS analogues can eventually be used as guidelines for isolating R genes in pistachio (Bahramnejad, 2014). LEAFY homologous gene (PcBLFY) of P. chinensis, which is the key regulator of flower development, was cloned and its sequence was analysed (Zhang et al., 2012). BLAST analysis showed 89–99% similarity between PcBLFY and the corresponding regions of LEAFY and its orthologues. PcBLFY could be used for breeding cultivars with shorter juvenile phase and earlier flowering.
